En İyi Oyun Tony Ödülü Broadway'de sergilenen oyunlar için verilen tiyatro ödülü.
Tony Ödülleri'nin ilk yılında en iyi oyun için ödül verilmedi. Çoğunlukla All My Sons'ın 1947'de En İyi Oyun ödülü aldığı sanılır ama gerçekte o sene Arthur Miller'a En İyi Yazar ödülü verilmişti. Bir sonraki sene Mister Roberts En İyi Oyun Tony ödülünü alan ilk yapım oldu.
Hem yazar hem de yapımcılara ödül verilmektedir.
